目的:观察心肌细胞织反常时细胞内的游离钙浓度的变化,探讨丹参素和SOD对细胞内的游离钙浓度的影响。方法:体外分离成年大鼠心肌细胞复制氧反常模型,缺氧40min后给氧20min,用钙荧光探针(Fluo-3)分别测定了正常大鼠心肌细胞(Control,n=11)、缺氧40min(H40min,n=11)以及缺氧40min再给氧20min(H40minR20min,n=12)的大鼠心肌细胞内的游局钙浓度,并观察了丹参素(H40minR20min+DS,n=10)和SOD(H40minR20min+SOD,n=11)对氧反常的心肌细胞内的游离钙浓度的影响,并用显微镜观察了细胞杆形率的改变。结果:氧反常组的细胞内的游离钙浓度(596.71±20.29nmol/L)明显比正常对照和单纯缺氧40min的高(158.67±5.01和328,51±13.86nmol/LP<0.01)。加入丹参素和SOD,可明显降低氧反常细胞内的游离钙的浓度(分别为245.29±8.83和300.04±13.04nmol/L,P<0.01)。细胞杆形率亦有类似的改变。结论:氧反常时心肌细胞发生明显的钙超载,丹参素和SOD对细胞内钙超载具有一定的防治作用。
Objective: To observe the changes of intracellular free calcium concentration in cardiomyocytes abnormal cells and to explore the effect of danshensu and SOD on intracellular free calcium concentration. Methods: The anoxia model of adult rat cardiomyocytes was isolated in vitro. Oxygen was given to the rats for 40 min after oxygen deprivation for 20 min. Fluorescent calcium (Fluo-3) was used to measure the normal rat cardiomyocytes (Control, n = 11) (H40min, n = 11), and oxygenation 40min and then given oxygen 20min (H40minR20min, n = 12) in rat cardiomyocytes calcium concentration and observation of the Danshensu (H40minR20min + DS, n = 10) and SOD H40minR20min + SOD, n = 11) on the level of free calcium in cardiomyocytes with abnormal oxygen, and observed the change of rod shape by microscope. Results: The intracellular free calcium concentration (596.71 ± 20.29nmol / L) in the oxygen abnormal group was significantly higher than that in the normal control group and the hypoxia 40min group (158.67 ± 5.01 and 328,51 ± 13.86nmol /LP<0.01). The addition of Danshensu and SOD could significantly reduce the concentration of free calcium in oxygen-abnormal cells (245.29 ± 8.83 and 300.04 ± 13.04 nmol / L, P <0.01). Similar changes in cell rod shape. CONCLUSION: Significant calcium overload occurs in cardiomyocytes during anoxia. Danshensu and SOD have some preventive and therapeutic effects on intracellular calcium overload.
