目的为提高卫生监督机构对温泉浴水质的现场快速检测能力和手段,建立温泉浴池水中细菌总数新的检测方法。方法对温泉浴池水中非细菌细胞的ATP用生物方法分离,使用荧光虫素和荧火虫光素酶稀释溶剂将溶解的细菌细胞中的ATP转化为光能,再通过灵敏的微光度计检出ATP的量,通过查表,得出池水中所含细菌总数。结果该方法分离效果良好,重现性好,灵敏度、精密度高,干扰少。结论本方法携带、操作方便,重现性好,测定结果可靠,灵敏度高,无论在温泉浴池水还是游泳池水的现场快速检测都有较好的适用性。
Objective To improve the health supervision agencies on the hot spring water quality on-site rapid detection capabilities and means to establish a new total number of bacteria in hot spring water bath detection methods. Methods The ATP of non-bacterial cells in hot-spring bath water was isolated biologically, and the ATP in the solubilized bacterial cells was converted to light energy using a fluorogenic insecticide and a firefly luciferase enzyme, which was then detected by a sensitive microphotometer The amount of ATP, through the table, draw the total number of bacteria contained in the pool water. Results The method has good separation effect, good reproducibility, high sensitivity, high precision and less interference. Conclusion The method is portable, easy to operate, reproducible, reliable, sensitive and has good applicability in the field of hot spring bath or swimming pool water.
