为研究活血潜阳胶囊对血管紧张素Ⅱ (ATⅡ )诱导的血管平滑肌细胞 (VSMC)增殖的影响。以贴块法行SD大鼠胸主动脉VSMC分离培养 ,以血清药理学方法进行实验 ,10 - 6 MATⅡ为刺激因子 ,Captopril为对照 ,活血潜阳胶囊药物血清分为 10 %、7 5 %、5 %、2 5 % 4个浓度。绘制细胞生长曲线 ,并测定DNA的合成。结果 :经活血潜阳胶囊处理后 ,细胞增殖速度明显下降 ,生长曲线下移 ,3H -TdR掺入量减少 ,DNA合成明显受到抑制 ,且在一定范围内有剂量依赖性。提示 :活血潜阳胶囊能抑制ATⅡ引起的VSMC增生 ,揭示了活血潜阳胶囊治疗高血压的机理之一。
To investigate the effect of Huoxue Qianyang Capsule on the proliferation of vascular smooth muscle cells (VSMC) induced by angiotensin II (ATII). The rat thoracic aorta VSMC were isolated and cultured by the patch method. Serum pharmacology was used for the experiment. 10-6 MATII was the stimulatory factor, Captopril was the control, and Huoxue Qianyang capsule was divided into 10% and 75%. 5 %, 25 % 4 concentrations. Draw the cell growth curve and measure the DNA synthesis. RESULTS: After treated with Huoxue Qianyang capsule, the cell proliferation rate decreased significantly, the growth curve shifted downwards, 3H-TdR incorporation was decreased, DNA synthesis was significantly inhibited, and it was dose-dependent within a certain range. Tip: Huoxue Qianyang capsule can inhibit the proliferation of VSMC caused by ATII, revealing one of the mechanisms of Huoxue Qianyang Capsule in the treatment of hypertension.
