目的:观察熏洗I号对大鼠肛瘘术后模型创面表皮细胞生长因子(EGF)表达的影响。方法:SD大鼠40只制成肛瘘术后创面模型,并按随机数表达法分为生理盐水组(10只),创面用生理盐水熏洗;高锰酸钾组(10只),创面用高锰酸钾熏洗;实验组(10只),创面用自拟中药“熏洗I号”熏洗;正常组(10只)。于造模成功后第10天采用Western blot蛋白印迹及免疫组化法观察EGF在创面的表达,作为创面肉芽组织增生情况的指标;造模成功后第1天、第10天从大鼠眼底静脉丛采血检测血液学变化,作为炎症变化指标;测定大鼠创面愈合率,作为判断创面愈合情况的指标;采用光密度法测定4组大鼠EGF的表达量。结果:以愈合率达到95%为创面愈合,实验组创面平均愈合时间15 d,与其它3组对比具有统计学差异。血液学结果提示在术后10 d中药熏洗I号组大鼠的炎症表现得到明显控制。Western blot蛋白印迹及免疫组化法显示,实验组大鼠创面肉芽组中EGF的阳性表达明显高于其它3组(P<0.01)。结论:自制中药洗剂“熏洗I号”可以促进肛周污染性创面释放表皮细胞生长因子(EGF),加速上皮化进程,从而达到促进创面愈合的作用。
Objective: To observe the effects of fumigation I on the expression of epidermal cell growth factor (EGF) in rat models of postoperative anal fistula. Methods: Forty SD rats were divided into three groups: normal saline group (n = 10) and saline group (n = 10). The wounds were treated with potassium permanganate Potassium permanganate fumigation; experimental group (10), wound with self-made traditional Chinese medicine “fumigation I number” fumigation; normal group (10). Western blotting and immunohistochemistry were used to observe the expression of EGF on the wound surface on the 10th day after successful modeling as an indicator of wound granulation tissue hyperplasia. On the first day and the tenth day after modeling, The hemorrhagic changes of blood samples were collected and the hemorrhagic changes were detected as the index of inflammation changes. The wound healing rate was determined as the index to judge the healing status of wounds. The expression of EGF in four groups of rats was measured by optical density. Results: With healing rate of 95% as wound healing, the average wound healing time of the experimental group was 15 days, which was statistically different from the other three groups. The results of hematology showed that the inflammation of rats in Group I was obviously controlled after 10 days of operation. Western blot and immunohistochemistry showed that the positive expression of EGF in wound granulation group was significantly higher than that in other three groups (P <0.01). Conclusion: The self-made Chinese medicine lotion “Fumigation I” can promote perianal contaminating wound to release epidermal cell growth factor (EGF) and accelerate the process of epithelization, so as to promote wound healing.
