目的观察活脉通络饮对脑缺血再灌注大鼠Bcl-2、Bax蛋白表达的影响,探讨活脉通络饮的神经保护机制。方法 Wistar大鼠随机分为正常对照组、缺血再灌注组、活脉通络饮低剂量组、活脉通络饮中剂量组、活脉通络饮高剂量组和银杏叶片阳性对照组。采用改良的线栓法,建立大鼠大脑中动脉栓塞(MCAO)脑缺血再灌注模型,给予大鼠评定神经功能缺损程度评分,采用免疫组织化学技术和WesternBlot检测脑组织中Bcl-2、Bax的表达。结果模型组大鼠脑组织Bax蛋白的表达较正常对照组明显增加(P<0.01),给予活脉通络饮治疗后,Bax蛋白表达与模型组相比明显减少(P<0.01),而各组的Bcl-2蛋白表达无明显变化(P>0.05)。结论活脉通络饮的脑保护作用机制之一可能与下调Bax蛋白表达相关。
Objective To observe the effect of active maitongluoyin on the expression of Bcl-2 and Bax in rats with cerebral ischemia-reperfusion, and to explore the neuroprotective mechanism of active maitonggluoin. Methods Wistar rats were randomly divided into normal control group, ischemia-reperfusion group, low-dose Huoxue Tongluo Yin group, middle-dose Huoxue Tongluo Yin group, high-dose Huoxue Tongluo decoction group and positive control group of Ginkgo biloba leaves. A rat model of middle cerebral artery occlusion (MCAO) cerebral ischemia-reperfusion was established by a modified thread embolism method. Rats were assessed for neurological impairment scores. Bcl-2 and Bax in brain tissue were detected by immunohistochemistry and Western Blot. expression. Results The expression of Bax in the brain of the model group was significantly higher than that in the normal control group (P<0.01). After the treatment of Huomong Tongluo, the expression of Bax protein was significantly reduced compared with the model group (P<0.01). There was no significant change in Bcl-2 protein expression in the group (P>0.05). Conclusion One of the mechanisms of brain protective effect of Huomai Tongluo Yin may be related to the down-regulation of Bax protein expression.
