本研究用4例成人尸体延髓,年龄为19~45岁,死后2~4 h内取材。先用自来水与蒸馏水冲洗,继而以4%多聚甲醛与0.2%戊二醛0.1 mol/L磷酸缓冲液浸泡,固定8~12 h,再移入10~20%蔗糖磷酸缓冲液中10 h,冰冻切片,厚40μm。将切片分为4套,1套做对照试验,其余3套分别以兔抗5羟色胺(5-hydroxytryptamine,5-HT)、亮脑啡肽(leucine-enkephalin,L-ENK)和生长抑素(so-matostatin,SOM)特异血清做免疫组织化学反应。
