目的探讨ING4及CD34蛋白在人前列腺癌组织中的表达及相关性。方法采用免疫组织化学Elivsion TM Plus二步法检测28例人前列腺癌及32例良性前列腺增生组织中ING4及CD34的表达情况,分析其结果与前列腺癌病理分级及危险等级关系。结果 ING4基因蛋白在人前列腺癌组织中的阳性表达率明显低于良性前列腺增生组织,且随肿瘤病理分级、PSA及临床分期的增加而降低。而CD34标记的微血管密度(microvessle density,MVD)在前列腺癌组织中表达明显较良性前列腺增生组织高,其MVD值随肿瘤病理分级及危险等级增加而升高。前列腺癌组织中ING4表达与MVD值呈负相关,r值为-0.827,相关性具有统计学意义(P<0.05)。结论 ING4基因蛋白在人前列腺癌组织表达异常,可能参与了肿瘤的发生及进展,同时可以作为前列腺癌临床评价诊断、指导治疗的辅助指标之一。
Objective To investigate the expression and correlation of ING4 and CD34 protein in human prostate cancer. Methods The expressions of ING4 and CD34 in 28 cases of human prostate cancer and 32 cases of benign prostatic hyperplasia were detected by immunohistochemistry Elivsion TM Plus two-step method. The relationship between the expression of ING4 and CD34 was analyzed and the pathological grade and risk grade of prostate cancer were analyzed. Results The positive rate of ING4 protein in human prostate cancer was significantly lower than that in benign prostatic hyperplasia (BPH), and decreased with the increase of PSA and clinical stage. However, the MVD of CD34-labeled microvessel density (MVD) was significantly higher in benign prostatic hyperplasia tissues than that in benign prostatic hyperplasia tissues. The MVD value increased with the increase of tumor grade and risk grade. The expression of ING4 in prostate cancer tissues was negatively correlated with MVD, with a r value of -0.827. The correlation was statistically significant (P <0.05). Conclusions The expression of ING4 protein in human prostate cancer tissues may be involved in the occurrence and progression of tumors, and may serve as an adjunct to clinical diagnosis of prostate cancer.
