流感病毒RNA聚合酶对流感病毒基因组的复制和表达至关重要。编码聚合酶各亚基的基因序列高度保守的特点使其成为极具发展潜力的抗流感药物靶标。本研究通过构建流感病毒RNA聚合酶活性依赖的荧光素酶报告系统,在细胞水平上建立了以流感病毒RNA聚合酶为靶点的抗流感药物筛选模型。特异性评价和统计学分析表明,该筛选方法灵敏可靠、重复性好,可以用于针对流感病毒RNA聚合酶的抗流感药物筛选。
Influenza virus RNA polymerase is essential for the replication and expression of the influenza virus genome. The highly conserved nature of the gene sequences encoding each subunit of the polymerase makes it a promising anti-influenza drug target. In this study, influenza virus RNA polymerase-dependent luciferase reporter system was constructed to establish an influenza-resistant RNA polymerase-based drug screening model at the cellular level. Specificity evaluation and statistical analysis showed that the screening method is sensitive and reproducible, and can be used for anti-influenza drug screening of influenza virus RNA polymerase.
