为观察牛主动脉中蛋白聚糖对培养的人脐静脉内皮细胞生长的影响及其在动脉粥样硬化形成中的作用,用解聚提取、离子交换及凝胶过滤柱层析法从牛主动脉内、中膜分离出硫酸乙酰肝素蛋白聚糖、硫酸软骨素蛋白聚糖和硫酸皮肤素-硫酸软骨素蛋白聚糖。用氚标胸腺嘧啶脱氧核苷掺入法和细胞计数法观察上述三种蛋白聚糖对培养的第一代人脐静脉内皮细胞生长的影响。结果发现,用氚标胸腺嘧啶脱氧核苷掺入法计算硫酸乙酰肝素蛋白聚糖的抑制率(醛酸含量2.1~8.3mg/L)分别为28.3%、52.3%和75.4%,硫酸软骨素蛋白聚糖的抑制率(醛酸含量9.5~37.8mg/L)分别为61%、83%和91%,硫酸皮肤素-硫酸软骨素蛋白聚糖的抑制率(醛酸含量9.3~36.9mg/L)分别为58.4%、82.4%和86%。用细胞计数法计算硫酸乙酰肝素蛋白聚糖(醛酸含量2.1~8.3mg/L)的抑制率分别为21.5%、37.8%、62.9%,硫酸软骨素蛋白聚糖(醛酸含量9.5~37.8mg/L)的抑制率分别为37.2%、68.9%和76.5%,硫酸皮肤素硫酸软骨素蛋白聚糖(醛酸含量9.3~36.9mg/L)的抑制率分别为44.5%、61.6%和71.7%。结果?
