目的探讨永生化人支气管上皮细胞(human bronchial epithelial cells,BEAS-2B)端粒保卫蛋白1(protection of telomere 1,POT1)的表达下调与煤焦沥青(coal tar pitch,CTP)致端粒损伤的作用。方法 BEAS-2B细胞以CTP组,二甲基亚砜(dimethylsulfoxide,DMSO)溶剂对照组和空白对照组分别进行培养。培养至1代、10代、20代和30代时分别对细胞进行POT1基因RNA干扰,然后进行端粒长度、POT1和人端粒酶逆转录酶(human telomer-ase reverse transcriptase,hTERT)的mRNA及蛋白表达情况的检测。结果与对照组相比,抑制POT1基因的表达可导致hTERT的mRNA表达水平下调(F=25.008,P<0.001),端粒DNA相对长度缩短(F=14.617,P=0.001),随着培养代数的增加,端粒DNA相对长度缩短(F=15.988,P<0.001)。结论抑制POT1基因表达可间接促进CTP对BASE-2B细胞的端粒损伤作用。
OBJECTIVE: To investigate the effect of down-regulation of telomere 1 (POT1) expression in immortalized human bronchial epithelial cells (BEAS-2B) on telomere damage induced by coal tar pitch (CTP) effect. Methods BEAS-2B cells were cultured in CTP group, dimethylsulfoxide (DMSO) solvent control group and blank control group respectively. After 1, 10, 20 and 30 passages of culture, the cells were respectively subjected to RNA interference of POT1 gene, followed by telomere length, mRNA of POT1 and human telomer-ase reverse transcriptase (hTERT) And protein expression detection. Results Compared with the control group, inhibition of POT1 gene expression resulted in a down-regulation of mRNA expression of hTERT (F = 25.008, P <0.001) and shorter relative length of telomere DNA (F = 14.617, P = 0.001) The relative length of telomere DNA shortened (F = 15.988, P <0.001). CONCLUSION: Inhibition of POT1 gene expression can indirectly promote the telomere injury of BASE-2B cells induced by CTP.
