胡椒酸己二胺对高脂血症大鼠糖脂代谢的影响

目的:研究胡椒酸己二胺(PAH)对高脂饲料诱导大鼠糖脂代谢的影响。方法:雄性SD大鼠78只,适应性饲养一周后,测定基础血清胆固醇(TC)和甘油三酯(TG),随机抽取12只作为正常对照组,给予普通饲料,余作为造模组喂以高脂饲料。喂养4周后,再次检血清TC、TG、低密度脂蛋白胆固醇(LDL-C),比较造模前后血脂水平,TC、TG、LDL-C显著升高,认为造模成功。将造模成功的60只大鼠按血脂大小分层,并随机分为5组,即高脂模型组、辛伐他汀组(10mg/kg)和胡椒酸己二胺10mg/kg、5mg/kg、2.5mg/kg 3个剂量组,每日灌胃给予各受试物。第4周末处理动物,采集空腹血清,测定TC、TG、HDL-C、LDL-C、空腹血糖(FPG)及空腹胰岛素(FINS)水平,同时制作石蜡切片,光镜观察各组大鼠肝脏组织形态变化。结果:(1)胡椒酸己二胺能显著降低高脂血症大鼠的血清TC、TG和LDL-C水平,且呈剂量依赖性;2.5mg/kg剂量时显著升高HDL-C水平;降血脂作用与传统降血脂药物辛伐他汀相比,以降LDL-C作用更为明显。(2)胡椒酸己二胺也能显著降低高脂血症大鼠FPG水平。(3)与高脂模型组比较,胡椒酸己二胺5mg/kg剂量可降低胰岛素抵抗指数(HOMA-IR),提示胡椒酸己二胺具有改善胰岛素抵抗作用。(4)胡椒酸己二胺5mg/kg剂量组明显改善肝细胞肿胀情况,对肝细胞有一定的保护作用。结论:胡椒酸己二胺具有降低血脂、血糖,改善胰岛素抵抗的作用。 Objective: To study the effect of captopril (PAH) on glucose and lipid metabolism in rats induced by high fat diet. Methods: Seventy-eight male Sprague-Dawley rats were fed with normal diet for one week. Baseline serum cholesterol (TC) and triglyceride (TG) were measured. Twelve were randomly selected as normal control group. High-fat feed. Four weeks after feeding, serum TC, TG and LDL-C were tested again. Blood lipid levels before and after modeling were significantly increased, and TC, TG and LDL-C were significantly increased. 60 successful rats were stratified according to the size of blood lipids and randomly divided into 5 groups: high fat model group, simvastatin group (10mg / kg) and hexamethylenediamine hexamine 10mg / kg, 5mg / kg , 2.5mg / kg 3 dose groups, given to each subject by daily gavage. At the end of the fourth week, the animals were collected and the fasting serum was collected. The levels of TC, TG, HDL-C, FPG and FINS were determined. Paraffin sections were made and the liver tissues Morphological changes. Results: (1) Hexamethylenediamine hexahydrate significantly reduced the serum levels of TC, TG and LDL-C in hyperlipidemic rats in a dose-dependent manner; HDL-C levels were significantly increased at 2.5mg / kg; Hypolipidemic effect compared with traditional lipid-lowering drugs simvastatin, in order to reduce the role of LDL-C more obvious. (2) Capsicum also inhibited FPG level in hyperlipidemic rats. (3) Compared with the high-fat model group, the dose of 5mg / kg hexamethylenediamine decreased the insulin resistance index (HOMA-IR), suggesting that hexamethylenediamine hexamine could improve the insulin resistance. (4) The hexamine 5 mg / kg dose group significantly improved hepatocyte swelling and had some protective effect on hepatocytes. Conclusion: Hexamethylenediamine hexamine has the effect of lowering blood fat, blood sugar and improving insulin resistance.
