目的 :比较益肾壮骨、健脾利湿和养血柔肝中药对原发 OA黑鼠关节软骨内几种细胞因子基因转录水平的影响。方法 :以运动负荷造成 C5 7黑鼠 OA模型 ,以 RT- PCR方法测定各组动物关节软骨内 IL- 1β、IL- 6、i NOS m RNA基因转录水平。结果 :C5 7黑鼠关节软骨 IL- 1β、i NOS m RNA表达随增龄逐渐增强 ,OA中期最明显 ,晚期下降。在 OA早、中期 ,益肾方可轻度下调关节软骨 IL- 1β、i NOS的基因表达 ,对 IL- 6有轻度上调作用 ;健脾方和柔肝方可明显下调 IL- 1β、i NOS的基因表达 ,与芬必得比较 ,有显著性差异 ,对 IL- 6无明显影响。结论 :不同治则方药对关节软骨细胞因子的基因表达影响不一 ;健脾方和柔肝方可明显下调鼠关节软骨 IL - 1、i NOSm RNA表达。
Objective : To compare the effect of Yishen Zhuanggu, Jianpi Lishi and Yangxue Rougan on the transcription of several cytokine genes in the articular cartilage of primary OA rats. METHODS: The C5 7 black rat OA model was induced by exercise load. The transcription levels of IL-1β, IL-6, and iNOS mRNA in the articular cartilage of each group were determined by RT-PCR. Results : The expression of IL-1β and iNOS mRNA in the articular cartilage of C57 mice gradually increased with increasing age. In the early and middle stage of OA, Yishen prescription can mildly down-regulate the expression of IL-1β and iNOS gene in articular cartilage, and slightly up-regulate IL-6. Jianpifang and Rougan recipe can significantly down-regulate IL-1β and i. The gene expression of NOS was significantly different from that of Fenbid, and had no significant effect on IL-6. Conclusion : Different treatment prescriptions have different effects on gene expression of articular chondrocyte cytokines; Jianpi Fanghe Rougan Recipe can significantly down-regulate IL-1, iNOSm RNA expression in rat articular cartilage.
