胆固醇由甾体母核和侧链构成,是一种新的甾体资源.以胆固醇为唯一碳源的选择培养基,利用富集培养法从土壤中筛选出一株细菌,编号5901.该菌能够快速高效转化胆固醇,专一性生成4-胆甾烯-3-酮.经形态学观察和16S rDNA序列比对分析,该菌株为Rhodococcus sp.该菌株转化胆固醇的最优条件为:生长温度和转化温度均为30℃,初始pH 8.0.胆固醇质量浓度为5 g/L,反应24 h后,胆固醇转化率为95%.本研究表明,590 1菌株较之前报道的菌株,底物浓度高,转化速率快,也可以催化其它甾体化合物的3β-羟基氧化和5位双键位移反应,具有潜在的研究和应用价值.
