目的研究羊水预处理在羊水细胞培养瓶法中的应用效果。方法采用羊水预处理培养瓶法(实验组)和未经处理羊水细胞培养瓶法(对照组)对100例18-24周的孕中期孕妇和152例25-32周的孕晚期孕妇进行羊水细胞培养。结果 100例孕18-24周羊水细胞培养实验组培养成功率高于对照组(P<0.05),实验组羊水细胞培养时间比对照组短(P<0.001),实验组有效核分裂数比对照组多(P<0.001);152例孕25-32周羊水细胞培养实验组培养成功率高于对照组(P<0.01),实验组羊水细胞培养时间比对照组短(P<0.001),实验组有效核分裂数比对照组多(P<0.001)。结论羊水预处理羊水细胞培养瓶法羊水细胞培养成功率高,细胞培养时间短,有效核分裂数多,可进行羊水产前诊断孕妇孕周范围增宽。
Objective To study the application of amniotic fluid preconditioning in amniotic fluid culture flask method. Methods A total of 100 pregnant women of 18-24 weeks of pregnancy and 152 pregnant women of 25-32 weeks of pregnancy were treated with amniotic fluid pretreatment culture flask method (experimental group) and untreated amniotic fluid culture flask method (control group) to cultivate. Results The success rate of culture in amniotic fluid cell culture group was higher than that in control group (P <0.05). The culture time of amniotic fluid in experimental group was shorter than that in control group (P <0.001), and the effective mitosis in experimental group was higher than that in control group (P <0.001). The success rate of culture in amniotic fluid cell culture group was higher than that of control group (P <0.01) in 152 pregnant women with 25-32 weeks of pregnancy, and the experimental group was shorter than control group (P <0.001) The effective number of mitosis than the control group (P <0.001). Conclusion Amniotic fluid preconditioning with amniotic fluid culture flask method of amniotic fluid cell culture success rate, cell culture time is short, the effective number of mitosis, prenatal diagnosis of pregnant women can be broadened range of gestational weeks.
