目的探索北柴胡Bupleurum chinense侧根发育过程及影响侧根发育的因素,为北柴胡产量提升及商品性改良提供技术支撑。方法对北柴胡品种川北柴1号主根进行石蜡切片观察侧根原基发育动态图,以及添加不同浓度生长调节物质后进行根系参数考察。结果北柴胡侧根原基起源于中柱鞘细胞,部分内皮层细胞也参与侧根原基发育;施加10μmol/L IAA和Ca2+浓度均最大化促进北柴胡根系发育,而浓度过高则抑制侧根发育;施加0.1μmol/L TIBA和10μmol/L EDTA显著促进柴胡根系发育,浓度过高则起抑制效果。结论本研究方法还可以广泛应用于人参、当归等以根入药的药用植物中,对药用植物栽培及育种工作具有重大意义。
Objective To explore the process of lateral root development of Bupleurum chinense in North Bupleurum and its influencing factors on lateral root development to provide technical support for the yield increase and commercial improvement of Bupleurum chinense. Methods The dynamic changes of lateral root primordia were observed by paraffin sectioning of the main roots of Chuanbei Chai No.1 in North Chaihu, and the root parameters were investigated after adding different concentrations of growth regulators. Results Bupleurum root originated from pericycle cells and some endoderm cells were also involved in the development of lateral root primordium. The application of 10μmol / L IAA and Ca2 + both maximized the root development of N. bidentata. However, Development; Application of 0.1μmol / L TIBA and 10μmol / L EDTA significantly promoted the root system development of Bupleurum, but the effect was inhibited when the concentration was too high. Conclusion The method of this study can also be widely used in medicinal plants with ginseng, Chinese angelica and other medicinal plants rooted for medicinal purposes, which is of great significance to the cultivation and breeding of medicinal plants.
