目的测定中药戒毒康酊剂中氢溴酸东莨菪碱及总生物碱的含量。方法采用Diamonsil C18柱(250 mm×4.6 mm,5μm)分离测定氢溴酸东莨菪碱,流动相:以甲醇-0.02 mol.L-1醋酸钠溶液(含0.02%三乙胺和0.3%四氢呋喃,用冰醋酸调pH到6.2)梯度洗脱;流速1 mL.min-1;检测波长:206 nm。采用酸性染料比色法测定戒毒康中的总生物碱,0.2%溴百里香酚兰溶液显色,420 nm处测得吸光度。结果氢溴酸东莨菪碱在1.0~5.0μg内呈良好的线性关系(r=0.999 6);平均回收率为98.8%,RSD为2.29%(n=6)。总生物碱在0.04~0.2 mg内呈良好的线性关系(r=0.999 8);平均回收率为97.6%,RSD为2.06%(n=6)。结论本方法简便、快速、准确、专属性强,可作为戒毒康酊剂的检验方法。
Objective To determine the content of scopolamine hydrobromide and total alkaloids in traditional Chinese medicine Jie Du Kang tincture. Methods Scopolamine hydrobromide was separated and determined by Diamonsil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol-0.02 mol·L-1 sodium acetate solution containing 0.02% triethylamine and 0.3% Acetic acid to pH 6.2) gradient elution; flow rate 1 mL.min-1; detection wavelength: 206 nm. The total alkaloids in JiDuKang were determined by acid dye colorimetric method. The color was determined by 0.2% bromine thymol blue solution and the absorbance was measured at 420 nm. Results Scopolamine hydrobromide showed a good linearity (r = 0.999 6) in the range of 1.0-5.0 μg with an average recovery of 98.8% and a RSD of 2.29% (n = 6). The total alkaloids showed a good linear relationship in the range of 0.04-0.2 mg (r = 0.999 8). The average recovery was 97.6% and the RSD was 2.06% (n = 6). Conclusion The method is simple, rapid, accurate and specific, and can be used as a test method for drug treatment of tincture.
