目的:研究肺癌A549细胞和人胚肺成纤维细胞相互作用对细胞增殖及MMP-2表达的影响。方法:A549细胞与人胚肺成纤维细胞条件培养基共同培养,人胚肺成纤维细胞与A549细胞条件培养基共同培养,人胚肺成纤维细胞与不同比例A549细胞共同培养,分别用MTT和明胶酶谱法测定A549细胞和人胚肺成纤维细胞的细胞活力及MMP-2的表达。结果:培养24、48、72h后,人胚肺成纤维细胞条件培养基共同培养的A549细胞活力与同期对照组A549细胞活力相比,差异无统计学意义,P>0.05。A549细胞条件培养基共同培养的人胚肺成纤维细胞活力高于同期对照组A549细胞活力,差异有统计学意义,P=0.000。A549细胞条件培养基促进人胚肺成纤维细胞MMP-2酶原和活化酶的表达,人胚肺成纤维细胞条件培养基对A549细胞MMP-2的表达无影响。人胚肺成纤维细胞按不同比例与A549细胞共同培养时,48hMMP-2酶原活性和表达逐渐增强,且1∶8比例共培养时出现少量MMP-2活化酶,72h除1∶1比例培养时MMP-2酶原外,MMP-2酶原和活化酶活性和表达均增强。结论:肺癌A549细胞和人胚肺成纤维细胞相互作用能促进细胞增殖及MMP-2的表达和活化。
Objective: To investigate the effect of lung cancer A549 cells and human embryo lung fibroblast interaction on cell proliferation and MMP-2 expression. Methods: A549 cells were co-cultured with human embryonic lung fibroblast conditioned medium. Human embryonic lung fibroblasts were co-cultured with conditioned medium of A549 cells. Human embryonic lung fibroblasts were co-cultured with different proportions of A549 cells and cultured with MTT and Gelatin zymography was used to determine cell viability and MMP-2 expression in A549 cells and human embryonic lung fibroblasts. Results: The viability of A549 cells co-cultured with human embryonic lung fibroblast conditioned medium was not significantly different from that of A549 cells in control group at 24, 48 and 72 hours after culture (P> 0.05). The viability of human embryonic lung fibroblasts co-cultured with A549 cells conditioned medium was higher than that of A549 cells in the same period, the difference was statistically significant (P = 0.000). A549 cell conditioned medium promoted the expression of MMP-2 and activated enzyme in human embryo lung fibroblasts. The conditioned media of human embryonic lung fibroblasts had no effect on the expression of MMP-2 in A549 cells. When human embryonic lung fibroblasts were co-cultured with A549 cells in different proportions, the activity and expression of MMP-2 at 48h gradually increased. A small amount of MMP-2-activating enzyme appeared in co-culture at 1: When MMP-2 zymogen, MMP-2 zymogen and activase activity and expression were enhanced. Conclusion: The lung cancer A549 cells and human embryo lung fibroblasts interaction can promote cell proliferation and MMP-2 expression and activation.
