体外培养人脐静脉内皮细胞,观察拉米普利酸对由Cu2+、内皮细胞氧化修饰的LDL损伤内皮细胞的保护作用及与NO、cGMP的关系。结果发现:OLDL损伤内皮细胞,使MDA升高,NO、cGMP下降。拉米普利酸能有效对抗OLDL损伤内皮细胞的作用,降低MDA,升高NO、cGMP。一氧化氮合成酶拮抗剂硝基左旋精氨酸(N-L-Arg)能加强OLDL的损伤效应,且能部分拮抗拉米普利酸的保护作用。结果提示:拉米普利酸对OLDL损伤血管内皮细胞的保护作用与其促进内皮细胞NO合成和释放有关。
Human umbilical vein endothelial cells were cultured in vitro and the protective effect of Lamipril on endothelial cells oxidatively modified by Cu2 + and endothelial cells was observed and its relationship with NO and cGMP was observed. The results showed that: OLDL injured endothelial cells, increased MDA, NO, cGMP decreased. Ramipril can effectively antagonize the effect of OLDL on endothelial cells, reduce MDA, increase NO, cGMP. Nitric oxide synthase antagonist nitro-L-arginine (N-L-Arg) can enhance the damage effect of OLDL, and can partially antagonize the protective effect of Lamipril. The results suggest that the protective effect of Lamipril on endothelial cells injured by OLDL is related to the promotion of endothelial nitric oxide synthase (NO) synthesis and release.
