目的研究8周有氧运动对视黄醇结合蛋白4(RBP4)诱导的胰岛素抵抗鼠骨骼肌和肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物的影响,探讨运动缓解RBP4诱导的胰岛素抵抗机制。方法RBP4腹腔注射建立胰岛素抵抗鼠模型。设8周有氧运动组(RBP4 aerobic exercise group,RA)、安静对照组(RBP4control group,RC)和正常安静对照组(Control group,C)。检测血清RBP4、胰岛素、血糖,计算胰岛素抵抗指数HOMA-IR;RT-PCR法检测SOCS-3mRNA表达;免疫共沉淀法检测SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物表达。结果 (1)与C组相比,RC组小鼠血清RBP4、HOMA-IR均显著增加,骨骼肌、肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物均显著增加。(2)与RC组相比,RA组小鼠血清RBP4、HOMAIR均显著降低,骨骼肌、肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物均显著减少。结论 8周有氧运动能够显著改善RBP4诱导的胰岛素抵抗,机制涉及降低血清RBP4水平,减少高RBP4诱导的SOCS-3表达,以及SOCS-3与IR、IRS结合形成的复合物,恢复正常的胰岛素信号传导。
Objective To investigate the effect of 8-week aerobic exercise on the expression of SOCS-3mRNA, SOCS-3 and IR, IRS phosphorylated protein in skeletal muscle and liver induced by retinol binding protein 4 (RBP4) in rats Exercise attenuates RBP4-induced insulin resistance. Methods Rat model of insulin resistance was established by intraperitoneal injection of RBP4. RBP4 aerobic exercise group (RA), RBP4control group (RC) and normal control group (C) were established. Serum RBP4, insulin and blood glucose were measured to calculate HOMA-IR. SOCS-3 mRNA expression was detected by RT-PCR. The expression of SOCS-3 complex with IR and IRS phosphorylated protein was detected by co-immunoprecipitation. Results (1) Compared with group C, the serum RBP4 and HOMA-IR in RC group increased significantly, the SOCS-3 mRNA expression in skeletal muscle and liver, SOCS-3 complex with IR and IRS phosphorylated protein were significantly increase. (2) Compared with RC group, serum RBP4 and HOMAIR in RA group were significantly decreased, SOCS-3 mRNA expression in skeletal muscle and liver, SOCS-3 and IR, phosphorylated IRS protein complexes were significantly reduced. Conclusion Aerobic exercise at 8 weeks can significantly improve RBP4-induced insulin resistance by decreasing serum RBP4 level, decreasing high RBP4-induced SOCS-3 expression, and complex formed by the combination of SOCS-3 with IR and IRS, restoring normal insulin Signaling.
