目的制定甘草渣药材质量标准,为甘草渣的合理开发利用提供科学依据。方法采用薄层鉴别法(TLC)对甘草渣进行定性鉴别,参照《中国药典》方法进行常规项目检查,以甘草苷为对照品,采用紫外分光光度法测定甘草渣中总黄酮的含量,采用高效液相色谱法测定甘草渣中甘草酸和甘草苷的含量。结果甘草渣药材薄层色谱中,在对照品相同位置显相同颜色斑点。甘草渣中水分平均含量为7.66%,灰分平均含量为7.94%,酸不溶性灰分平均含量为2.17%。紫外分光光度法表明甘草苷回归方程为:y=5.462x-0.008 3(r=0.999 7),回收率为100.74%(RSD=1.38%),5批甘草渣中总黄酮平均含量为1.81%。HPLC定量分析,甘草苷和甘草酸的回归方程为y甘草苷=14 314x+31.44(r=0.999 2);y甘草酸=3892.9x+21.473(r=1)。结果表明,甘草苷在0.222~1.11 mg/m L浓度范围内,甘草酸在0.36~2.16 mg/m L浓度范围内,峰面积与浓度呈良好的线性关系,平均回收率分别为100.74%和99.34%,RSD分别为1.38%和2.50%。不同批次甘草苷和甘草酸平均含量为0.019%和0.57%。结论所建立的分析方法简便、准确、可靠,具有很好的专属性和准确性,可用于甘草渣药材的质量控制。
Objective To establish the quality standard of licorice residue medicinal materials and provide a scientific basis for the rational exploitation and utilization of licorice residue. Methods Qualitative identification of licorice residue was carried out by TLC. According to the method of “Chinese Pharmacopoeia”, routine test was conducted. The content of total flavonoids in licorice residue was determined by UV spectrophotometry. Determination of Glycyrrhizin and Glycyrrhizin in Licorice by Liquid Chromatography. Results licorice residue TLC, the same position in the reference substance was the same color spots. Licorice residue in the average moisture content of 7.66%, the average ash content of 7.94%, acid-insoluble ash average content of 2.17%. UV spectrophotometry showed that the regression equation of glycyrrhizin was: y = 5.462x-0.008 3 (r = 0.999 7), the recovery rate was 100.74% (RSD = 1.38%). The average content of total flavonoids in five batches of licorice residue was 1.81%. HPLC quantitative analysis of glycyrrhizin and glycyrrhizic acid regression equation for the y glycyrrhizin = 14 314x + 31.44 (r = 0.999 2); y glycyrrhizic acid = 3892.9x +21.473 (r = 1). The results showed that there was a good linear relationship between the concentration of glycyrrhizin and the concentration range of 0.36 ~ 2.16 mg / mL in the concentration range of 0.222 ~ 1.11 mg / mL, with the average recoveries of 100.74% and 99.34 %, RSD respectively 1.38% and 2.50%. Different batches of licorice glycyrrhizin and the average content of 0.019% and 0.57%. Conclusion The established analytical method is simple, accurate and reliable. It has good specificity and accuracy and can be used for the quality control of licorice residue.
