本文用高压液相方法测定茶碱药物浓度,观察了17例哮喘儿童一次静脉推注氨茶碱后的经时血药变化,用电子计算机自动求算出茶碱代谢动力学参数:消除半衰期为5.10±1.26h,分布容积为0.513±0.056l/kg,总清除率为74±20ml/h/kg。通过不同给药方法的稳态血药浓度水平及波动情况的观察,探讨了适合于我国儿童的茶碱使用方法。结果表明,口服普通氨茶碱片,应每6h服用5.5mg/kg(每天总量不超过600~800mg),可使用多数患儿血药水平达到有效治疗药物浓度。
In this paper, high-pressure liquid-phase method for theophylline drug concentration was observed in 17 asthmatic children after a single intravenous injection of aminophylline plasma drug changes, the computer automatically calculated theophylline metabolic kinetics parameters: elimination half-life of 5.10 ± 1.26h, the volume of distribution was 0.513 ± 0.056l / kg, the total clearance rate was 74 ± 20ml / h / kg. Through the different administration methods of steady-state blood drug levels and fluctuations in the observation of suitable for our children’s theophylline use. The results showed that oral administration of aminophylline tablets should be taken every 6h 5.5mg / kg (total daily dose of not more than 600 ~ 800mg), the majority of children can be used to achieve effective drug levels of blood drug levels.
