目的通过动物离体平滑肌实验测试分析系统,观察五加皮提取物对大鼠肠平滑肌的作用,并对其中的作用机制进行分析探讨。方法分析五加皮提取物对乙酰胆碱、Ca Cl2和高钾引起的鼠十二指肠平滑肌自主收缩的影响,并对其中的作用机制进行初步分析。结果五加皮提取物对鼠离体十二指肠平滑肌自主收缩、Ach诱导鼠十二指肠平滑肌收缩、高钾去极化诱导的鼠十二指肠平滑肌收缩均具有明显抑制作用,在0.8~4.0 mg·m L?1内呈明显的量效关系。结论五加抑制肠平滑肌收缩的机制可能与阿托品相似,即可能为M胆碱受体阻断剂或作用于其他受体操纵的Ca2+通道,与电压依赖性通道无关。
OBJECTIVE: To study the effect of the extract of Wujiapi on rat intestinal smooth muscle through the experimental test and analysis system of the isolated rat smooth muscle, and to explore the mechanism of action. Methods The effects of Wujianpi extract on autonomic contractility of rat duodenal smooth muscle induced by acetylcholine, CaCl 2 and potassium were analyzed and the mechanism of action was analyzed. Results Wujiapi extract on autologous contraction of isolated rat duodenal smooth muscle, Ach induced duodenal smooth muscle contraction, high potassium depolarization induced rat duodenal smooth muscle contraction were significantly inhibited at 0.8 ~ 4.0 mg · m L · 1 showed a significant dose-effect relationship. Conclusion The mechanism of pentagastrin in inhibiting intestinal smooth muscle contraction may be similar to that of atropine, which may be M cholinergic receptor blockers or Ca2 + channels acting on other receptors, independent of voltage-dependent channels.
