目的检测慢性乙型肝炎(CHB)患者外周血单个核细胞(PBMCs)中T细胞受体重排删除环(TRECs)的含量,进而评价CHB患者的胸腺近期输出功能,并分析TRECs含量与HBV病毒含量间的关系。方法利用实时荧光定量PCR法检测205例CHB患者PBMCs中TRECs含量及血清HBV DNA含量,同时选取32例健康人群作为对照组。结果患者组外周淋巴细胞中TRECs含量明显低于对照组,差异有统计学意义(P<0.05)。根据HBV DNA含量的不同分为HBV DNA阴性组、低病毒组(<1×105IU/ml)和高病毒组(≥1×105IU/ml),3组患者TRECs含量差异无统计学意义(t=2.58,P>0.05)。采用直线回归相关性分析发现,HBV DNA含量与TRECs含量之间无相关性(r=0.15,P>0.05)。结论 CHB患者PBMCs中TRECs含量低于正常人群,提示CHB患者胸腺近期输出功能明显降低;TRECs含量与HBV病毒含量间无明显相关性。
Objective To detect the content of T cell receptor rearrangement deletion loops (TRECs) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis B (CHB), and to evaluate the recent thymic output function of CHB patients. The relationship between content. Methods The contents of TRECs and serum HBV DNA in PBMCs of 205 CHB patients were detected by real-time fluorescence quantitative PCR. 32 healthy people were selected as the control group. Results The content of TRECs in peripheral lymphocytes of patients was significantly lower than that of control group (P <0.05). According to the content of HBV DNA, there were no significant differences in the content of TRECs between the three groups (HBV DNA negative group, low virus group (<1 × 105 IU / ml) and high virus group (≥1105 IU / ml) 2.58, P> 0.05). Linear regression analysis showed no correlation between HBV DNA content and TRECs content (r = 0.15, P> 0.05). Conclusions The content of TRECs in PBMCs of patients with CHB is lower than that in normal subjects, which indicates that the output function of thymus in patients with CHB is obviously decreased. There is no significant correlation between the content of TRECs and the content of HBV.
