目的观察益气活血化痰法对慢性阻塞性肺疾病(COPD)模型大鼠细胞因子及气道重塑的影响,探讨益气活血化痰法治疗COPD的作用机理。方法通过香烟熏结合脂多糖(LPS)气管滴入法,建立COPD大鼠模型。观察益气活血化痰方药对大鼠血清中TNF-α、IL-8和肺泡灌洗液中MUC5AC的表达水平及其对各级支气管和肺组织病理变化的影响。结果益气活血化痰方药中、高剂量组可显著降低COPD大鼠血清IL-8、TNF-α及BALF中MUC5AC的表达水平,其中,高剂量组效果优于中剂量组。结论益气活血化痰方药中、高剂量组能降低COPD大鼠血清中IL-8、TNF-α和BALF中MUC5AC的表达水平,抑制气道炎性反应和缓解气道重塑,揭示益气活血化痰法临床疗效可能的作用机制,为临床治疗提供科学依据。
Objective To observe the effect of Yiqi Huoxue Huatan method on cytokines and airway remodeling in chronic obstructive pulmonary disease (COPD) model rats, and to explore the mechanism of Yiqi Huoxue phlegm method in treating COPD. Methods COPD rat model was established by cigarette smoke combined with lipopolysaccharide (LPS) tracheal instillation. To observe the effect of Yiqi Huoxue Huatan Recipe on serum TNF-α, IL-8 and MUC5AC in bronchoalveolar lavage fluid and its effect on bronchial and lung histopathological changes at all levels. Results The Yiqi Huoxue Huatan recipe medium and high dose groups can significantly reduce the serum IL-8, TNF-α and the MUC5AC expression in BALF in COPD rats, of which, the high dose group is better than the middle dose group. Conclusion The Yiqi Huoxue Huatan recipe can reduce the levels of IL-8, TNF-α in serum and the expression of MUC5AC in BALF in COPD rats, inhibit the airway inflammatory reaction and relieve airway remodeling, Huoxue phlegm method of clinical efficacy of the possible mechanism for clinical treatment to provide a scientific basis.
