高迁移率蛋白A2在垂体腺瘤中的表达及意义

目的研究高迁移率蛋白A2(HMGA2)在垂体腺瘤中的表达及其与垂体腺瘤侵袭性的相关性。方法采用免疫组织化学法及RT-PCR法检测55例垂体腺瘤组织中HMGA2蛋白的表达。依据Wilson改良Hardy分类系统分级分期标准分组:侵袭性垂体腺瘤组18例,非侵袭性垂体腺瘤组37例。依据内分泌学检查分为:无功能性垂体腺瘤5例,功能性垂体腺瘤50例。结果 HMGA2在无功能垂体腺瘤中无表达,在功能性垂体腺瘤中表达率82%,差异有统计学意义。在侵袭性组表达阳性率32.43%,与侵袭性垂体腺瘤(100%)相比,差异有统计学意义。侵袭组HMGA2mRNA的表达水平较非侵袭组表达水平显著增高(0.891±0.172vs.0.372±0.181),功能性垂体瘤较非功能性垂体瘤HMGA2mRNA的表达水平显著增高(0.564±0.196vs.0),差异有统计学意义。结论 HMGA2的高表达在功能性垂体瘤的发生及垂体瘤的侵袭性中发挥重要的作用。 Objective To investigate the expression of HMGA2 in pituitary adenomas and its relationship with the invasiveness of pituitary adenomas. Methods The expression of HMGA2 protein in 55 cases of pituitary adenoma was detected by immunohistochemistry and RT-PCR. According to Wilson’s Modified Hardy classification system, the graded staging group consisted of 18 cases of invasive pituitary adenoma and 37 cases of non-invasive pituitary adenoma. According to endocrine examination are divided into: 5 cases of non-functional pituitary adenoma, 50 cases of functional pituitary adenoma. Results There was no expression of HMGA2 in non-functioning pituitary adenomas, and the positive rate of HMGA2 in functional pituitary adenomas was 82%. The difference was statistically significant. The positive expression rate in invasive group was 32.43%, which was significantly different from that in invasive pituitary adenoma (100%). The expression level of HMGA2 mRNA in invasive group was significantly higher than that in non-invasive group (0.891 ± 0.172 vs.0.372 ± 0.181). The expression of HMGA2 mRNA in functional pituitary tumor was significantly higher than that in non-functional pituitary tumor (0.564 ± 0.196 vs.0) The difference was statistically significant. Conclusion The high expression of HMGA2 plays an important role in the development of functional pituitary tumors and the invasiveness of pituitary tumors.
