目的:研究酪氨酸蛋白激酶抑制剂槲皮素(quercetin)对血小板衍生生长因子(PDGF)诱导的血管平滑肌细胞(SMC)酪氨酸磷酸化蛋白表达的影响。方法:采用噻唑蓝比色法,流式细胞术分析槲皮素对PDGF诱导的SMC增殖的影响,应用免疫印迹法分析观察槲皮素对PDGF诱导的SMC酪氨酸磷酸化蛋白表达的影响。结果:与对照组相比,槲皮素处理组可显著地抑制PDGF诱导的SMC增殖及酪氨酸磷酸化蛋白的表达,尤以酪氨酸磷酸化蛋白分子量为85~180kD明显。结论:槲皮素可显著地抑制PDGF诱导的SMC酪氨酸磷酸化蛋白的表达。
AIM: To investigate the effect of quercetin, a tyrosine kinase inhibitor, on tyrosine phosphorylation of vascular smooth muscle cells (SMC) induced by platelet-derived growth factor (PDGF). Methods: The effect of quercetin on the proliferation of SMC induced by PDGF was analyzed by flow cytometry. The effect of quercetin on the expression of SMC tyrosine phosphorylation protein induced by PDGF was observed by Western blotting. Results: Compared with the control group, the quercetin treatment group could significantly inhibit PDGF-induced SMC proliferation and tyrosine phosphorylation protein expression, especially tyrosine phosphorylated protein molecular weight of 85 ~ 180kD significantly. Conclusion: Quercetin can significantly inhibit PDGF-induced SMC tyrosine phosphorylation protein expression.
