目的:研究加减资生汤配方颗粒对肺气肿模型大鼠肺组织及血清IL-4、IL-8、TNF-α表达的影响。方法:采用烟熏结合气管注射脂多糖法造模,随机分为空白组、模型组、低剂量组、中剂量组、高剂量组,每天给药1次,连续给药10周。病理切片染色法观察肺气肿模型大鼠给药前后肺、支气管等组织的病理变化。采用PCR结合酶联免疫吸附法,检测大鼠肺组织、血清IL-4、IL-8、TNF-α表达水平。结果:与空白组相比,模型组大鼠肺、支气管、血清TNF-α、IL-8、IL-4表发水平显著增强(P<0.05);与模型组相比,加减资生汤配方颗粒干预组能显著降低肺气肿模型大鼠肺组织及血清TNF-α、IL-4、IL-8表达水平(P<0.05)。结论:加减资生汤干预肺气肿的机制可能与其调节肺组织及血清中TNF-α、IL-4、IL-8表达有关。
Objective: To study the effects of Zisheng decoction granules on the expression of IL-4, IL-8 and TNF-α in lung tissues and serum of rats with emphysema. Methods: The model of smoked and tracheal injection of lipopolysaccharide was established and randomly divided into blank group, model group, low dose group, medium dose group and high dose group, once a day for continuous administration for 10 weeks. Pathological sections were used to observe the pathological changes in lung, bronchus and other tissues before and after administration of emphysema rats. The expression of IL-4, IL-8 and TNF-α in the lung tissues and serum of rats were detected by PCR and enzyme-linked immunosorbent assay. Results: Compared with the blank group, the levels of TNF-α, IL-8 and IL-4 in the lung, bronchus and serum of the model group were significantly increased (P <0.05). Compared with the model group, The formula granule intervention group can significantly reduce the lung tissue and serum TNF-α, IL-4 and IL-8 expression levels in emphysema rats (P <0.05). Conclusion: The mechanism of interfering with emphysema may be related to regulating the expression of TNF-α, IL-4 and IL-8 in lung tissue and serum.
