目的 建立成年牛眼晶状体上皮细胞的培养方法 ,研究其在不同培养液中的生长特性并观察其超微结构。 方法 组织块静置贴壁培养 ,应用细胞计数法和噻唑蓝 (MTT)法检测细胞的生长情况 ,透射电镜观察所培养细胞的超微结构。 结果 晶状体上皮细胞在高糖和低糖的 DMEM培养液中的原代及传代培养生长均良好 ,在低糖 DMEM中能更快速贴壁 ;电镜观察可见细胞内微丝丰富 ,高尔基体发达 ,溶酶体多少不等 ,粗面内质网丰富和线粒体少。 结论 组织块静置培养是合适的晶状体上皮细胞体外培养方法。细胞生长状况与培养液中含糖量关系不大 ,其较强的贴壁能力可能与所含丰富的微丝有关。
Objective To establish a culture method of adult bovine lens epithelial cells and study its growth characteristics in different culture media and observe its ultrastructure. Methods The tissue explants were placed on stationary culture. The cell growth was measured by cytometry and MTT assay. The ultrastructure of cultured cells was observed by transmission electron microscopy. Results The primary and subculture cultures of lens epithelial cells in DMEM medium with high glucose and low glucose grew well and adhered more rapidly in DMEM with low glucose. Electron microscopy showed that intracellular micro-filaments were abundant, the Golgi body developed, lysosomes How many different, rich in rough endoplasmic reticulum and less mitochondria. Conclusion Tissue mass stationary culture is a suitable method for in vitro culture of lens epithelial cells. Cell growth conditions and the broth of sugar content has little to do, and its strong adherent ability may be related to the rich microfilament.
