本文报道5例骨肉瘤患者一次静脉滴注氨甲喋呤(MTX)的药代动力学研究。选用荧光分光光度法测定不同时间点的MTX血浆浓度,经计算机处理表明其体内过程呈二室开放模型并获得药动学参数。实验结果证明,为保证大剂量MTX——FC解救疗法的安全合理应用,有必要进行MTX的血药浓度监测。
This article reports the pharmacokinetics of a single intravenous infusion of methotrexate (MTX) in 5 patients with osteosarcoma. Fluorescence spectrophotometry was used to determine the plasma concentration of MTX at different time points. The computerized treatment showed that the in vivo process was a two-compartment open model and pharmacokinetic parameters were obtained. The experimental results show that in order to ensure the safe and rational use of high dose MTX - FC rescue therapy, it is necessary to monitor the blood concentration of MTX.
