目的探讨反义黏着斑激酶(FAK)对U251人胶质瘤细胞株生物学行为影响。方法以LipofecTAMINE~(TM)介导的反义FAK寡核苷酸转染U251人胶质瘤细胞株,用Western blot检测胶质瘤细胞株FAK含量的改变,研究其多种细胞生物学行为的变化。结果脂质体介导FAK ODN转染率为85.9%。转染反义FAK后,U251人胶质瘤细胞株FAK表达明显减少,L1251细胞生长抑制率为64.52%,侵袭细胞下降至21.7个,细胞周期分析显示s期细胞降至25.16%,细胞凋亡率升至34.5%。透射电镜观察到凋亡细胞。结论反义FAK寡核苷酸可显著降低FAK在U251 中的表达,降低FAK表达能够显著抑制U251人胶质瘤细胞株侵袭能力,抑制细胞增殖和诱导凋亡。
Objective To investigate the effect of FAK on the biological behavior of U251 human glioma cell line. Methods U251 human glioma cell line was transfected with antisense FAK oligonucleotide mediated by LipofecTAMINE ™ and Western blot was used to detect the changes of FAK in glioma cell lines. Variety. Results Liposome-mediated FAK ODN transfection rate was 85.9%. After transfection with antisense FAK, the expression of FAK in U251 human glioma cell lines was significantly reduced, the growth inhibition rate of L1251 cells was 64.52%, and the invasion cells decreased to 21.7 cells. Cell cycle analysis showed that cells in s phase decreased to 25%. 16%, the apoptosis rate rose to 34.5%. Apoptotic cells were observed by transmission electron microscopy. Conclusion Antisense FAK oligodeoxynucleotides can significantly reduce the expression of FAK in U251. Decreasing the expression of FAK can significantly inhibit the invasiveness of U251 human glioma cell lines and inhibit cell proliferation and induce apoptosis.
