目的建立反相高效液相色谱-双波长切换法同时测定天舒胶囊中天麻素、阿魏酸和6,7-二羟基藁本内酯的含量。方法采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-体积分数0.1%乙酸水溶液为流动相,梯度洗脱,流速为1.0 mL.min-1,检测波长为270 nm(天麻素)、320 nm(阿魏酸和6,7-二羟基藁本内酯),柱温35℃。结果在15 min内天舒胶囊中天麻素、阿魏酸和6,7-二羟基藁本内酯质量浓度分别在30.50~305.0 mg.L-1(r=0.999 2)、34.96~349.6 mg.L-1(r=0.999 7)和46.54~465.4 mg.L-1(r=0.999 4)内与峰面积线性关系良好;平均加样回收率分别为98.8%、98.7%和98.2%,RSD分别为1.9%、1.2%和2.1%。结论本方法操作简便,结果准确,与《中华人民共和国药典》所述方法相比,增加了含量测定指标,提高了全面控制天舒胶囊质量的水平。
OBJECTIVE To establish a reversed-phase high performance liquid chromatography-dual wavelength switch method for the simultaneous determination of gastrodin, ferulic acid and 6,7-dihydroxy ligustilide in Tianshu capsules. Methods A Kromasil C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of methanol and 0.1% acetic acid aqueous solution with a gradient of 1.0 mL · min-1. The detection wavelength was 270 nm (gastrodin ), 320 nm (ferulic acid and 6,7-dihydroxy ligustilide) at 35 ° C. Results The concentration of gastrodin, ferulic acid and 6,7-dihydroxy-ligustilide in Tianshu capsules were 30.50 ~ 305.0 mg.L-1 (r = 0.999 2) and 34.96 ~ 349.6 mg in 15 min respectively. The linearity of peak areas was good in L-1 (r = 0.999 7) and 46.54-465.4 mg.L-1 (r = 0.999 4), respectively. The average recoveries were 98.8%, 98.7% and 98.2% 1.9%, 1.2% and 2.1%. Conclusion The method is easy to operate and the results are accurate. Compared with the method described in the “Pharmacopoeia of the People’s Republic of China”, this method increases the content determination index and improves the quality of Tianshu capsule.
