目的 :探讨幽门螺杆菌 (Hp)对低转移胃癌细胞生长和裸鼠接种转移的影响 ,并建立动物转移实验体外模型。方法 :应用细胞计数及MTT测定等方法 ,观察Hp活菌对胃癌细胞系BGC - 82 3低转移细胞株生长的影响。并行裸鼠接种 ,观察裸鼠转移情况。结果 :细胞计数及MTT检测结果均显示 :Hp活菌菌液浓度≥ 4 .0×10 5CFU/ml时 ,细胞生长明显受抑 ,且浓度越高抑制越显著 ,细胞存活率与Hp活菌的浓度呈负相关 ,与对照组相比均有显著性差异 (P <0 .0 0 1)。裸鼠接种实验提示实验组肿瘤大小与对照组比较的差异 (P <0 .0 5 )。病理结果示实验组 2只裸鼠发现转移灶 ,而对照组无转移。结论 :低Hp活菌浓度能促进BGC - 82 3细胞增殖 ,且可能加速肿瘤的生长与转移
Objective: To investigate the effect of Helicobacter pylori (Hp) on the growth of low metastatic gastric cancer cells and the inoculation and metastasis of nude mice and to establish an animal model of in vitro metastasis. Methods: The effects of Hp viable cells on the growth of gastric cancer cell line BGC - 82 3 were observed by cell counting and MTT assay. Parallel nude mice were inoculated to observe the metastasis of nude mice. Results: The results of cell counting and MTT showed that the cell growth was significantly inhibited when the concentration of Hp viable bacteria was ≥ 4 × 10 5 CFU / ml, and the higher the concentration was, the more obvious the inhibition was. The cell survival rate was similar to that of Hp Concentration was negatively correlated, compared with the control group were significantly different (P <0. 001). Inoculation experiments in nude mice suggested that the tumor size in the experimental group was significantly different from that in the control group (P <0.05). Pathological results showed that two nude mice in experimental group found metastases, but no metastasis in control group. CONCLUSION: Low Hp viability can promote the proliferation of BGC - 82 3 cells and accelerate the growth and metastasis of tumor
