目的探讨氯化镧(LaCl_3)暴露对仔鼠海马紧密连接蛋白表达和基因转录水平影响。方法将40只Wistar孕鼠随机分为对照组(蒸馏水)和0.25%、0.50%、1.00%(w/v)氯化镧组,从仔鼠出生起孕鼠通过饮水染毒LaCl_3,断乳后仔鼠继续饮用原浓度LaCl_3溶液1个月;采用Western blot法检测仔鼠海马claudin-5、occludin和紧密连接蛋白-1(ZO-1)蛋白表达,real-time PCR法检测仔鼠海马Cldn5、Ocln和Tjp1 mRNA表达。结果与对照组比较,0.50%和1.00%氯化镧组仔鼠海马claudin-5蛋白表达水平[分别为(0.059±0.007)、(0.041±0.003)]与occludin蛋白表达水平[分别为(0.085±0.004)、(0.067±0.004)]均明显降低(P<0.05);1.00%氯化镧组仔鼠海马Cldn5、Ocln和Tjp1 mRNA表达水平分别为(0.743±0.026)、(0.900±0.119)、(0.931±0.100),与对照组比较,1.00%氯化镧组仔鼠海马Cldn5 mRNA表达下降(P<0.05)。结论生长发育期暴露氯化镧可下调仔鼠海马claudin-5和occludin表达,导致血脑屏障紧密连接受损。
Objective To investigate the effect of lanthanum chloride (LaCl_3) exposure on the expression of hippocampal tight junction protein and gene transcription in neonatal rats. Methods Forty Wistar pregnant rats were randomly divided into control group (distilled water) and 0.25%, 0.50%, 1.00% (w / v) lanthanum chloride group. The pups continued to drink the original concentration of LaCl_3 solution for 1 month. Western blot was used to detect the expression of claudin-5, occludin and ZO-1 in the hippocampus of pups. Real-time PCR was used to detect the expression of Cldn5, Ocln and Tjp1 mRNA expression. Results Compared with the control group, the expression levels of claudin-5 protein in hippocampus of 0.50% and 1.00% lanthanum chloride-treated mice [(0.059 ± 0.007) and (0.041 ± 0.003), respectively] and occludin protein expression levels were (0.085 ± (0.673 ± 0.026), (0.900 ± 0.119) and (0.900 ± 0.119) respectively in the hippocampus of 1.00% Lanthanum chloride group (P <0.05). The mRNA expression levels of Cldn5, Ocln and Tjp1 in the hippocampus of 1.00% 0.931 ± 0.100). Compared with the control group, the expression of Cldn5 mRNA in the hippocampus of 1.00% Lanthanum chloride group decreased (P <0.05). Conclusion Exposure to lanthanum chloride during growth and development can down-regulate the expression of claudin-5 and occludin in the hippocampus of neonatal rats, resulting in the impaired adhesion of the blood-brain barrier.
