[目的]研究补脾益气方药对脾虚哮喘大鼠IL-13含量及IKKα表达的影响,揭示其平喘的作用机制。[方法] Wistar大鼠40只,分五组:对照组、脾虚哮喘组、哮喘组、脾虚哮喘中药组、哮喘中药组。ELISA检测大鼠支气管肺泡灌洗液(BALF)中1L-13含量;HE染色观察嗜酸性粒细胞(EOs)浸润情况;免疫组化观察肺组织中IKKα蛋白表达。[结果]与对照组比较,哮喘组和脾虚哮喘组IKKα表达及IL-13含量显著增高(P<0.01),且脾虚哮喘组IKKα表达和IL-13含量均高于哮喘组(P<0.05),中药治疗组和哮喘两组比较,IKKα表达和IL-13含量均降低(P<0.05)。[结论]补脾益气方药能调控脾虚哮喘大鼠肺组织中IKKα的表达及BALF中IL-13的含量。
[Objective] To investigate the effect of Bupi Yiqi Recipe on IL-13 content and IKKα expression in asthmatic rats with spleen asthenia and to reveal its mechanism of asthma. [Methods] Forty Wistar rats were randomly divided into five groups: control group, spleen asthma group, asthma group, spleen deficiency asthma TCM group and asthma TCM group. The content of 1L-13 in bronchoalveolar lavage fluid (BALF) was detected by ELISA. The infiltration of eosinophils (EOs) was observed by HE staining. The expression of IKKα in lung tissue was detected by immunohistochemistry. [Results] Compared with the control group, the expression of IKKα and the content of IL-13 in asthmatic group and spleen asthmatic group were significantly increased (P <0.01), and the levels of IKKα and IL-13 in asthmatic group were higher than those in asthmatic group (P <0.05) Compared with the asthma group, the expression of IKKα and the content of IL-13 in both Chinese medicine treatment group and asthma group were decreased (P <0.05). [Conclusion] BuBu Yi Qi Fang can regulate the expression of IKKα in lung tissue and the content of IL-13 in BALF in asthmatic rats with spleen asthma.
