目的对口岸截获的陈旧鼠样进行物种鉴定。方法运用DNA条形码技术对深圳盐田口岸入境,来自津巴布韦的集装箱内截获的腐败风干的鼠类残骸进行物种鉴定。分别从鼠眼周皮毛、腿部肌肉提取基因组DNA进行COI部分序列扩增,将扩增产物测序并比对,确定该样品物种。运用Mega 5.05软件构建系统发育树。结果经COI基因序列比对鉴定,截获鼠样为黑家鼠(Rattus rattus),与日本黑家鼠(GenBank号:AB752806)的Kimura 2-parameter遗传距离最近,值为0.003。结论 DNA条形码技术可以用于陈旧鼠样本的物种鉴定。
Objective Species identification of old mice intercepted at the port. Methods DNA barcoding was used to identify species of corrupted and dried rodents captured from containers in Zimbabwe at Shenzhen Yantian Port. The genomic DNA was extracted from the skin and muscle of the mouse eye respectively for COI partial sequence amplification. The amplified products were sequenced and compared to determine the sample species. Use Mega 5.05 software to construct phylogenetic tree. Results The sequence of COI gene was identified and the genetic distance of the intercepted Rattus rattus to the Kimura 2-parameter of Japanese black house mouse (GenBank accession number AB752806) was the closest, with a value of 0.003. Conclusion DNA barcoding technique can be used to identify species in old mice.
