目的比较不同训练方式对大鼠骨骼肌线粒体通透性转换孔(MPTP)及细胞凋亡的影响。方法以大鼠游泳训练和一次性力竭游泳为运动模型,将24只SD大鼠随机分为对照组(C)、游泳训练组(T)和一次性游泳力竭组(E)。T组大鼠进行无负重游泳训练6周,每周6次,1~2周每次训练45min,3~4周每次60min,5~6周为90min,末次训练24h后取股四头肌;E组正常饲养6周后于一次性力竭游泳后即刻取股四头肌。用紫外分光光度仪检测MPTP的开放;RT-PCR检测Bcl-2和Bax mRNA的表达;western blot检测细胞色素c(Cytc)的释放。结果①游泳训练没有导致大鼠骨骼肌MPTP的开放发生显著性变化,Bcl-2mRNA表达明显增强,Bax mRNA的表达及Cytc的释放量显著减少。②一次性力竭游泳导致大鼠骨骼肌MPTP的开放程度显著增大,Bcl-2mRNA表达显著降低,Bax mRNA表达和Cytc释放量显著增加。结论不同训练方式会通过调节骨骼肌线粒体通透性转换孔的开放及凋亡蛋白的释放来调节细胞凋亡。
Objective To compare the effects of different training methods on mitochondrial permeability transition pore (MPTP) and apoptosis in skeletal muscle of rats. Methods Totally 24 SD rats were randomly divided into control group (C), swimming training group (T) and disposable swimming exhaustion group (E). The rats in T group were trained on non-weight-bearing swimming for 6 weeks, 6 times per week, 45 minutes per session for 1 ~ 2 weeks, 60 minutes per 3 ~ 4 weeks, 90 minutes after 5 ~ 6 weeks, ; E group normal feeding six weeks after a one-time exhaustive swimming immediately after quadriceps. The opening of MPTP was detected by UV spectrophotometer. The expression of Bcl-2 and Bax mRNA was detected by RT-PCR. The release of cytochrome c (Cytc) was detected by western blot. Results ① Swimming training did not lead to a significant change in MPTP opening in skeletal muscle of rats. The expression of Bcl-2 mRNA was significantly increased, and the expression of Bax mRNA and the release of Cytc were significantly decreased. ② One-time exhaustive swimming resulted in a significant increase in the openness of MPTP in rat skeletal muscle, a significant decrease in Bcl-2 mRNA expression and a significant increase in Bax mRNA and Cytc release. Conclusion Different training methods regulate apoptosis by regulating the opening of mitochondrial permeability transition pore and the release of apoptotic protein.
