目的:研究蛇葡萄素对人胃癌细胞(SGC-7901)凋亡的影响。方法:MTT比色法测定细胞的增殖抑制率,流式细胞仪Annexin-V FITC/PI双染检测蛇葡萄素33.3,50,75 mg.L-1作用下SGC-7901细胞的凋亡及各细胞周期。结果:蛇葡萄素作用于SGC-7901 48 h后,可明显促进细胞早期凋亡,50,75 mg.L-1细胞凋亡率为(29.03±6.21)%,(43.43±1.00)%,明显高于空白组(11.83±4.67)%(P<0.05,P<0.01)且呈剂量依赖关系。蛇葡萄素33.3,50,75 mg.L-1作用于SGC-7901细胞48 h后,G1期细胞分别为(52.07±4.41)%,(57.07±2.70)%,(58.43±2.66)%明显高于空白对照组(42.00±3.04)%(P<0.05,P<0.01,P<0.01)。结论:蛇葡萄素能诱导SGC-7901细胞凋亡并使细胞周期阻滞在G1期。
Objective: To study the effect of snakegrass on the apoptosis of human gastric cancer cell line SGC-7901. Methods: MTT assay was used to determine the proliferation inhibition rate of SGC-7901 cells and the apoptotic rate of SGC-7901 cells was detected by flow cytometry Annexin-V FITC / PI double staining cell cycle. Results: After pretreatment with SGC-7901 for 48 h, the apoptosis rate of SGC-7901 cells was significantly increased (29.03 ± 6.21)% and (43.43 ± 1.00)%, respectively (11.83 ± 4.67)% (P <0.05, P <0.01) in a dose-dependent manner. The percentage of cells in G1 phase was (52.07 ± 4.41)%, (57.07 ± 2.70)%, (58.43 ± 2.66)%, respectively in SGC-7901 cells after 33.3,50,75 mg.L-1 treatment for 48 h In the blank control group (42.00 ± 3.04)% (P <0.05, P <0.01, P <0.01). Conclusion: Snakegrass can induce apoptosis of SGC-7901 cells and arrest the cell cycle in G1 phase.
