目的:探讨同种异体骨髓间充质干细胞局部多点注射联合特有中药膏剂外敷治疗兔糖尿病慢性溃疡创面治疗效果及结缔组织生长因子(connective tissue growth factor,CTGF)局部表达。方法:静脉注射四氧嘧啶(ALX)制备糖尿病兔模型,取建模成功的40只糖尿病兔,喂养3周后将各组动物后肢2 cm×3 cm全厚皮切除形成创面,随机数字表法分4组:A组:同种异体骨髓间充质干细胞多点联合特有中药膏剂治疗组(中药+干细胞组),B组:特有中药膏剂治疗组(中药组),C组:单独应用同种异体骨髓间充质干细胞治疗组(干细胞组),D组:糖尿病对照组(对照组)。各组治疗后于第7、14、21天分别观察创面面积;第21天取创面组织制备组织切片行Masson染色观察;应用Real Time PCR法检测创面组织中CTGF目的基因相对表达量。结果与结论:各治疗组创面愈合率明显高于对照组(P<0.01),A组和B组目的基因mRNA表达都上调,说明A组和B组治疗效果好,明显优于C,D组。
Objective: To investigate the therapeutic effect and the local expression of connective tissue growth factor (CTGF) in the treatment of chronic diabetic ulcer wounds by topical injection of allogeneic bone marrow mesenchymal stem cells combined with topical injection of Chinese traditional medicine. Methods: Diabetic rabbit models were established by injecting alloxan (IV) intravenously and 40 diabetic rabbits with successful modeling were sacrificed. After 3 weeks of feeding, 2 cm × 3 cm full thickness skin of hind limbs of all animals were resected to form wounds. Random number table Group A: allogeneic bone marrow mesenchymal stem cells multi-point combination of traditional Chinese medicine ointment treatment group (Chinese medicine + stem cell group), B group: the unique Chinese medicine treatment group (Chinese medicine group), C group: Allogeneic bone marrow-derived mesenchymal stem cells treated group (stem cell group), D group: diabetic control group (control group). The area of the wounds was observed on the 7th, 14th and 21st day after the treatment. On the 21st day, the tissue sections of the wound tissues were observed by Masson staining. The relative expression of CTGF in wound tissues was detected by Real Time PCR. RESULTS AND CONCLUSION: The wound healing rate of each treatment group was significantly higher than that of the control group (P <0.01), and the mRNA expression of the target genes was increased in both groups A and B, indicating that the treatment effect was better in group A and group B than in group C and D .
