目的:探讨丹参酮ⅡA对骨肉瘤MG-63细胞迁移、侵袭和基质金属蛋白酶(MMP-2、MMP-9)表达的影响。方法:不同浓度的丹参酮ⅡA处理MG-63细胞,细胞划痕法检测MG-63细胞迁移能力;Transwell小室体外侵袭实验法检测MG-63细胞侵袭能力;实时定量PCR法和蛋白印迹法(Western-blot)分别检测MG-63细胞金属基质蛋白酶(MMP-2/9)的mRNA及蛋白水平。结果:丹参酮ⅡA处理后,MG-63迁移明显下降;丹参酮ⅡA浓度在0,10,20,40μmol/L时穿过的MG-63细胞数分别为225.5±5.5、127.5±2.5、97.5±3.5、82.5±4.2个,MG-63细胞侵袭能力明显降低(P<0.05);丹参酮ⅡA处理明显降低MMP-2及MMP-9的mRNA和蛋白表达水平(P<0.05),且具有时间和浓度依赖性。结论:丹参酮ⅡA抑制骨肉瘤MG-63细胞的体外迁移和侵袭能力,这与降低基质金属蛋白酶的表达有关。
Objective: To investigate the effect of Tanshinone ⅡA on migration, invasion and expression of matrix metalloproteinase (MMP-2, MMP-9) in osteosarcoma MG-63 cells. Methods: The MG-63 cells were treated with different concentrations of tanshinone ⅡA, the migration ability of MG-63 cells was detected by cell scratch assay, the invasion ability of MG-63 cells was detected by Transwell chamber invasion assay in vitro, the real-time quantitative PCR and Western- blot was used to detect the mRNA and protein level of MMP-2/9 in MG-63 cells. Results: After Tanshinone Ⅱ A treatment, the migration of MG-63 significantly decreased. The number of MG-63 cells passing through Tanshinone ⅡA at 0, 10, 20 and 40 μmol / L were 225.5 ± 5.5, 127.5 ± 2.5 and 97.5 ± 3.5, (P <0.05). Tanshinone IIA treatment significantly decreased the mRNA and protein expression of MMP-2 and MMP-9 (P <0.05), and had a time-and concentration-dependent manner . Conclusion: Tanshinone Ⅱ A can inhibit the in vitro migration and invasion of osteosarcoma MG-63 cells, which is related to the decrease of the expression of matrix metalloproteinase.
