[目的]检测LMO1、Bcl-2、Bax、Survivin、Caspase-3蛋白在胃癌组织中的表达,并对这些蛋白之间的关系和临床意义进行分析。[方法]对96例胃癌手术患者的肿瘤组织标本及50例癌旁组织标本进行免疫组化染色(SP法),检测LMO1、Bcl-2、Bax、Survivin、Caspase-3蛋白的表达情况。同时收集患者的病理学情况,对与各指标的关系进行分析。[结果]胃癌组织中LMO1、Bcl-2、Survivin表达均比癌旁组织增强(均P<0.05),Caspase-3蛋白在胃癌组织中表达低于癌旁组织(P<0.05),Bax在两种组织中表达无明显差异(P>0.05)。胃癌组织中细胞凋亡率明显低于癌旁组织(P<0.01)。胃癌组织中LOM1蛋白强表达者细胞凋亡率明显低于弱表达者(P<0.01)。Spearman相关分析发现,胃癌组织中LMO1与Bcl-2表达存在正相关(r=0.3058,P=0.002),LMO1与Caspase-3表达存在负相关(r=-0.2886,P=0.004);Bcl-2与Survivin表达存在正相关关系(r=0.2986,P=0.0315),Bcl-2与Bax表达呈负相关(r=-0.4062,P<0.001),Survivin与Caspase-3也呈负相关(r=-0.3142,P=0.002)。[结论 ]胃癌组织中LMO1蛋白可能通过调节Bcl-2、Caspase-3的表达和活性而参与肿瘤凋亡。
[Objective] To detect the expression of LMO1, Bcl-2, Bax, Survivin and Caspase-3 protein in gastric cancer tissues, and to analyze the relationship between these proteins and their clinical significance. [Methods] Immunohistochemical staining (SP method) was performed on 96 specimens of gastric cancer patients and 50 paracancerous tissue specimens to detect the expression of LMO1, Bcl-2, Bax, Survivin and Caspase-3 proteins. At the same time, the patient’s pathology was collected and the relationship with each index was analyzed. [Results] The expressions of LMO1, Bcl-2 and Survivin in gastric cancer tissues were all stronger than those in adjacent tissues (all P<0.05). The expression of Caspase-3 protein in gastric cancer tissues was lower than that in adjacent tissues (P<0.05). There was no significant difference in the expression of the tissues (P>0.05). The rate of apoptosis in gastric cancer tissues was significantly lower than that in adjacent tissues (P<0.01). The apoptosis rate of gastric cancer tissues with strong LOM1 protein expression was significantly lower than that of weakly expressed ones (P<0.01). Spearman correlation analysis found that there was a positive correlation between LMO1 and Bcl-2 expression in gastric cancer tissues (r=0.3058, P=0.002). There was a negative correlation between LMO1 and Caspase-3 expression (r=-0.2886, P=0.004); Bcl-2 There was a positive correlation between Survivin expression (r=0.2986, P=0.0315), negative correlation between Bcl-2 and Bax expression (r=-0.4062, P<0.001), and negative correlation between Survivin and Caspase-3 (r=- 0.3142, P=0.002). [Conclusion] LMO1 protein may participate in tumor apoptosis by regulating the expression and activity of Bcl-2 and Caspase-3.
