目的观察MKK7/JNK途径对HCT116细胞恶性生长的影响。方法建立稳定敲低MKK7与JNK的细胞株,Western印迹实验检测JNK与MKK7的表达,集落形成技术检测HCT116细胞体外增殖。通过裸鼠荷瘤实验分析稳定敲低MKK7与JNK细胞株体内恶性生长能力。结果稳定敲低MKK7与JNK细胞株集落形成能力显著降低,成瘤能力明显减弱。结论 MKK7/JNK途径促进了HCT116细胞恶性生长。
Objective To observe the effect of MKK7 / JNK pathway on the malignant growth of HCT116 cells. Methods The cell lines stably knocked down MKK7 and JNK were established. The expression of JNK and MKK7 was detected by Western blotting. The proliferation of HCT116 cells was detected by colony formation assay. The malignant growth ability of MKK7 and JNK cell lines was stably knocked down by nude mice tumor-bearing experiment. Results Stable knockdown of MKK7 and JNK cell lines significantly reduced colony-forming ability, tumorigenic ability was significantly weakened. Conclusion MKK7 / JNK pathway promotes the malignant growth of HCT116 cells.
