目的:观察知母皂甙元(ZMS)对原代培养神经细胞M受体刺激后cGMP生成速率的影响。方法:用放射免疫分析法测定细胞内cGMP水平,观察ZMS不同作用时间对Carbachol刺激下cGMP生成的影响。结果:ZMS短时间(5min)作用于原代培养的神经细胞对Carbachol刺激下cGMP的生成无影响,而长时间(48h)作用明显提高Carbachol的刺激下cGMP生成量。结论:ZMS的作用与激动剂或拮抗剂不同,需要较长时间的作用才能发挥作用。
OBJECTIVE: To observe the effect of timosaponin (ZMS) on the rate of cGMP production in primary cultured neurons after M receptor stimulation. Methods: The intracellular cGMP levels were determined by radioimmunoassay. The effect of different time of ZMS on the cGMP production stimulated by Carbachol was observed. RESULTS: The short-term (5 min) administration of ZMS to primary cultured neurons had no effect on the production of cGMP by stimulation with Carbachol, but the long-term (48 h) effect significantly increased the stimulated cGMP production by Carbachol. Conclusion: The effect of ZMS is different from that of agonists or antagonists, and it takes a longer time to work.
