目的:采用浊点萃取法富集大鼠血浆中的双氯芬酸钠,并选择高效液相色谱法测定其在大鼠体内的血药浓度。方法:选用Triton X-114作为提取溶剂,色谱柱为DiamonsilTM C18色谱柱,检测波长225nm,流动相为乙腈-0.02mol·L-1醋酸钠溶液(35∶65,v/v)。结果:该方法在0.25~200μg·mL-1范围内线性良好,在低、中、高(0.5,25,160μg·mL-1)3个浓度的日内精密度RSD不大于7.7%,日间精密度RSD不大于11.4%。方法准确度RE在±5.8%范围内。3个浓度提取回收率不低于83.4%。结论:本方法简便、快捷、环保,适用于双氯芬酸钠在大鼠体内血药浓度的测定。
OBJECTIVE: To enrich the concentration of diclofenac sodium in rat plasma by cloud point extraction and to determine the concentration of diclofenac in rat plasma by high performance liquid chromatography. Methods: Triton X-114 was used as the extraction solvent. The column was DiamonsilTM C18 with a detection wavelength of 225 nm and a mobile phase of acetonitrile-0.02 mol·L-1 sodium acetate solution (35:65, v / v). Results: The method showed good linearity in the range of 0.25 ~ 200μg · mL-1. The intra-day RSD of three concentrations of low, medium and high concentrations (0.5,25,160μg · mL-1) was no more than 7.7% RSD not more than 11.4%. The method accuracy RE is within ± 5.8%. The recoveries of the three concentrations were not less than 83.4%. Conclusion: The method is simple, rapid and environmentally friendly and is suitable for the determination of diclofenac sodium in rats.
