目的 探讨化疗药物顺铂 (CDDP)对口腔鳞癌细胞凋亡及细胞周期特异性的诱导作用。方法 选用人舌鳞状细胞癌细胞系Tca8113为研究对象 ,应用倒置显微镜、荧光显微镜、透射电镜、流式细胞仪等方法进行观察。结果 CDDP作用Tca8113细胞后 ,细胞增殖变慢 ,增殖指数下降 ;细胞变小、变圆 ,失去贴壁性 ,从附着处脱落。DNA染色后荧光显微镜下观察 ,细胞核呈现橙红色、半月形、不规则块状等凋亡形态变化。透射电镜观察 ,细胞固缩 ,染色质浓聚边缘化 ,凋亡小体形成 ;流式细胞仪测定显示细胞周期发生明显改变 ,细胞分裂阻滞在S期。结论 CDDP能诱导Tca8113细胞凋亡 ,其作用有时间依赖性 ;诱导细胞凋亡可能是CDDP抗口腔鳞癌的一个重要机制
Objective To investigate the effect of chemotherapeutic drug cisplatin (CDDP) on apoptosis and cell cycle specificity of oral squamous cell carcinoma. Methods The human tongue squamous cell carcinoma cell line Tca8113 was selected as the research object, and observed by inverted microscope, fluorescence microscope, transmission electron microscope and flow cytometry. Results After CDDP treated Tca8113 cells, the proliferation of the cells was slower and the proliferation index decreased. The cells became smaller and rounder, and they lost adherence and fell off from the attachment. After staining with DNA, the nucleus was observed to be orange-red, half-moon, irregular, and other apoptotic morphological changes. Transmission electron microscopy showed that the cells were pyknosis, chromatin thickening was marginalized, and apoptotic bodies were formed. Flow cytometry showed that the cell cycle changed significantly, and cell division was arrested in S phase. Conclusion CDDP can induce apoptosis of Tca8113 cells in a time-dependent manner. Inducing apoptosis may be an important mechanism of CDDP against oral squamous cell carcinoma.
