目的:研究血小板第4因子(platelet factor 4,PF4)对用5.0 Gy60COγ照射损伤小鼠骨髓细胞凋亡的影响及其作用机制。方法:30只雄性BALB/c小鼠随机分为3组:正常对照组(6只)、PF4处理组(12只)和照射组(12只)。在照射前26、20 h分别给PF4处理组小鼠腹腔注射40μg/kg的PF4,再用5.0 Gy的60COγ射线全身均匀照射。照射后4、20h应用流式细胞术(FCM)测定小鼠骨髓细胞的凋亡率;并用Western blot检测凋亡相关蛋白Bax的表达。结果:照射后4、20 h PF4处理组的小鼠骨髓细胞的凋亡率低于照射组,两组比较差异有统计学意义(P<0.01)。Western blot检测显示照射后4、20 h照射组小鼠骨髓细胞Bax的表达要比正常对照组和PF4处理组的表达要高。结论:PF4可以抑制辐射所致小鼠骨髓细胞的凋亡,此保护机制可能与促凋亡蛋白Bax表达的下调有关。
Objective: To investigate the effect of platelet factor 4 (PF4) on the apoptosis of bone marrow cells of mice with radiation injury induced by 5.0 Gy60COγ and its mechanism. Methods: Thirty male BALB / c mice were randomly divided into three groups: normal control group (6 mice), PF4 treatment group (12 mice) and irradiation group (12 mice). PF4-treated mice were intraperitoneally injected with 40 μg / kg of PF4 at 26 and 20 h before irradiation, respectively, and then were uniformly irradiated with 5.0 Gy of 60 COγ rays. The apoptosis rate of mouse bone marrow cells was measured by flow cytometry (FCM) 4 and 20 hours after irradiation. The expression of apoptosis related protein Bax was detected by Western blot. Results: The apoptotic rates of bone marrow cells in PF4-treated mice at 4 and 20 h after irradiation were lower than those in irradiated mice, with significant difference between the two groups (P <0.01). Western blot showed that the expression of Bax in bone marrow cells of irradiated mice was higher than that of normal control group and PF4-treated group at 4 and 20 h after irradiation. Conclusion: PF4 can inhibit the apoptosis of mouse bone marrow cells induced by radiation, and the protective mechanism may be related to the down-regulation of the pro-apoptotic protein Bax.
