目的:观察胎盘多肽注射液对家兔骨折模型骨形态发生蛋白-2(BMP-2)及血管内皮生长因子(VEGF)表达的影响。方法:选用30只雄性新西兰家兔,随机分为3组,其中正常组10只,模型组10只,治疗组10只。正常组不予任何处理,模型组和治疗组均手术处理,制作骨折模型,治疗组家兔术后第6天开始每天注射胎盘多肽注射液4mL,所有家兔于4周后处死,免疫组织化学染色观察骨折部位骨形态发生蛋白-2(BMP-2)、血管内皮生长因子(VEGF)表达。结果:治疗组及模型组中BMP-2、VEGF表达量均高于正常对照组,且治疗组VEGF、BMP-2切片阳性细胞数高于模型组。结论:胎盘多肽注射液能提高骨折愈合过程中骨痂组织的BMP-2、VEGF表达。
Objective: To observe the effect of placental peptide injection on the expression of bone morphogenetic protein-2 (BMP-2) and vascular endothelial growth factor (VEGF) in rabbit fracture models. Methods: Thirty male New Zealand rabbits were randomly divided into three groups: normal group (n = 10), model group (n = 10) and treatment group (n = 10). The normal group received no treatment. The model group and the treatment group were surgically treated to make the fracture model. The rabbits in the treatment group were injected with 4 mL of the placental peptide injection daily on the 6th day after operation, and all the rabbits were sacrificed after 4 weeks. Immunohistochemistry The expression of bone morphogenetic protein-2 (BMP-2) and vascular endothelial growth factor (VEGF) in fracture site was observed by staining. Results: The expression of BMP-2 and VEGF in treatment group and model group were higher than those in normal control group, and the number of VEGF and BMP-2 positive cells in treatment group was higher than that in model group. Conclusion: Placental peptide injection can improve the expression of BMP-2 and VEGF in callus tissue during fracture healing.
