顺-1-环己基-2-羟甲基环己醇(6)经Anelli氧化和Pinnick氧化制得顺-1-羟基-(1,1’-二环己基)-2-甲酸(7),经(±)-α-苯乙胺拆分得异构体7a(1R,2R)和7b(1S,2S);另用D-樟脑酸和L-二苯甲酰酒石酸拆分1-二乙胺基-2-丙醇(8)得异构体8a(R)和8b(S)。7a/7b与8a/8b分别进行缩合,得罗西维林的4个单一异构体。
