目的研究超临界流体色谱对齐留通对映体的分离效果。方法考察确定5种手性柱中分离效果最佳的柱子,在该柱上考察改性剂的种类、浓度、温度及背压对分离度的影响。结果采用Chiralpak AD-H柱、改性剂为异丙醇,异丙醇在流动相中比例为18%、背压为140 bar、温度为303 K时,齐留通对映体达到最佳分离,分离度为11.5,出峰时间分别为4.59、7.35min。结论所用超临界流体色谱的方法简单、快捷、经济有效,适用于齐留通对映体的分离。
OBJECTIVE To study the separation effect of enzuron enantiomers by supercritical fluid chromatography. Methods The column with the best separation performance among the five chiral columns was investigated. The type, concentration, temperature and influence of the modifier on the separation were investigated. Results The Chiralpak AD-H column was used with isopropanol as the modifier. When isopropanol was present in the mobile phase at a ratio of 18%, back pressure of 140 bar and temperature of 303 K, the enantiomers of zileuton were optimally separated , Resolution of 11.5, the peak time was 4.59,7.35min. Conclusion The method of supercritical fluid chromatography is simple, rapid and cost-effective, and is suitable for the separation of enantiomers of zileuton.
