目的:观察治糖保肾冲剂对早期糖尿病肾病(diabetic nephropathy,DN)大鼠肾皮质转化生长因子(transforming growth factor β_1,TGFβ_1)表达的影响。方法:48只 Wistar 大鼠,分为中药(治糖保肾冲剂)组、西药(苯那普利)组、模型组及空白组共4组,每组各12只。采用多次腹腔注射链脲佐菌素(streptozo-tocin,STZ)建立早期 DN 大鼠模型,喂药4周。运用逆转录聚合酶链反应(RT-PCR)和免疫组化法重点观察DN 大鼠肾皮质 TGFβ_1蛋白、基因表达的影响。结果:造模各组肾皮质 TGFβ_1 mRNA 明显高于空白组,以模型组最高,中药组最低。中、西药组与模型组比较,中、西药组组间比较差异均有显著性(P<0.01)。各组肾皮质 TGFβ_1免疫组化比较,肾小管表达强于肾小球,组间排列:模型>西药>中药>空白。结论:益气活血补肾的治糖保肾冲刺治疗 DN 机理之一是抑制早期 DN 肾皮质 TGFβ_1蛋白、基因的过度表达。
OBJECTIVE: To observe the effect of Zhi Tang Bao Shen Granule on the expression of transforming growth factor β (TGFβ 1) in renal cortex of rats with early diabetic nephropathy (DN). Methods: Forty-eight Wistar rats were divided into four groups: traditional Chinese medicine (Zhitangbaoren) group, western medicine (benazepril) group, model group and blank group, 12 in each group. An early DN rat model was established by multiple intraperitoneal injections of streptozotocin (STZ) and administered for 4 weeks. Reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry were used to observe the effect of TGFβ_1 protein and gene expression in renal cortex of DN rats. Results: The expression of TGFβ_1 mRNA in the renal cortex was significantly higher in the model group than in the blank group, with the highest in the model group and lowest in the Chinese medicine group. Compared with the model group, there was significant difference between the Chinese medicine group and western medicine group (P<0.01). The renal cortex TGFβ_1 immunohistochemistry showed that the expression of renal tubules was stronger than that of the glomerulus. The arrangement between the groups was: model> Western medicine> Chinese medicine> blank. Conclusion: One of the mechanisms of the DN mechanism of Yiqi Huoxue Bushen treating glycoside-preserved kidney sprint is to inhibit the overexpression of TGFβ_1 protein and gene in the early DN renal cortex.
