目的:优选D101型大孔吸附树脂纯化鸡骨草总黄酮的工艺条件。方法:以芦丁为对照品,采用紫外比色法测定总黄酮含量,以吸附率和洗脱率为指标,通过单因素试验考察对大孔树脂动态吸附和解吸条件。结果:最佳工艺条件为上样质量浓度3 g.L-1,上样流速3 BV.h-1,2 BV 80%乙醇为洗脱剂,pH 6.0,洗脱流速3.5 BV.h-1。鸡骨草黄酮纯度由17.95%提高到52.80%。结论:D101型大孔树脂对鸡骨草黄酮类物质有较好的纯化效果,该优选的工艺简单可行。
Objective: To optimize the process conditions for purifying total flavonoids from Abrus cantoniensis with macroporous resin D101. Methods: Rutin was used as reference substance, and the content of total flavonoids was determined by UV colorimetric method. The adsorption and desorption conditions of macroporous resin were investigated by single factor test with adsorption rate and elution rate as indexes. Results: The optimum conditions were the loading mass concentration 3 g.L-1, the sample flow rate 3 BV.h-1,2 BV 80% ethanol as the eluent, pH 6.0 and the elution flow rate 3.5 BV.h-1. Clove flavonoids purity increased from 17.95% to 52.80%. Conclusion: The D101 macroporous resin has good purification effect on the flavonoids of Clove. The optimal process is simple and feasible.
